Neurons generated from induced pluripotent stem cells (iPSCs) from patients with Timothy syndrome showed abnormalities in differentiation compared with iPSC-derived neurons from controls, including decreased activity of genes that are expressed in lower cortical layers of the brain and in neurons that project to other areas of the cortex through the corpus callosum. Timothy syndrome–derived neurons also increased the production of norepinephrine and dopamine, which could be prevented by a calcium-channel blocker.
